Trials for preparation of hyperimmune serum against canine distemper virus conjugated with flourescine isothiocyanate

Abstract

Hyper – immune serum conjugated with flourescein isothiocyanate (FIT) against canine distemper virus (CDV) was prepared. Canine hyperimmunoglobulin against (CDV) was prepared in susceptible puppies. Immunoglobulin was separated from the blood serum by concentrated ammonium sulphate solution, then purified and conjugated with FIT. The non – specific fluorescence was removed from the conjugate using mice liver powder. The prepared CDV immunoglobulin conjugated with FIT was tested for its specificity using different smears of CDV antigen with control non inoculated smears. Clear positive reactions were obtained. The prepared conjugated hyperimmune serum gave a positive reaction up to a dilution of 1 : 32.
